me%22%3A%22Dirk%22%2C%22lastName%22%3A%22Vanderschueren%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Geert%22%2C%22lastName%22%3A%22Carmeliet%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Vanessa%22%2C%22lastName%22%3A%22Dubois%22%7D%2C%7B%22creatorType%22%3A%22author%22%2C%22firstName%22%3A%22Steve%22%2C%22lastName%22%3A%22Stegen%22%7D%5D%2C%22abstractNote%22%3A%22Transgender%20individuals%20are%20increasingly%20seeking%20gender-affirming%20therapy.%20For%20children%20and%20adolescents%2C%20this%20typically%20involves%20puberty%20suppression%20followed%20by%20hormone%20treatment.%20However%2C%20potential%20long-term%20adverse%20effects%20on%20the%20growing%20skeleton%20remain%20poorly%20characterized%2C%20largely%20due%20to%20the%20lack%20of%20appropriate%20preclinical%20animal%20models.%20Existing%20models%20often%20rely%20on%20surgical%20puberty%20suppression%20and%2C%20in%20the%20case%20of%20estradiol%20%28E2%29%20administration%2C%20cause%20a%20high%20bone%20mass-phenotype%20that%20is%20not%20observed%20in%20the%20clinical%20setting.To%20address%20this%2C%20we%20developed%20a%20novel%20preclinical%20mouse%20model%20that%20mimics%20the%20medical%20approach%20used%20in%20transgirls%20and%20analyzed%20the%20effects%20of%20gender-affirming%20therapy%20on%20bone.%20Four-week-old%20male%20mice%20were%20treated%20with%20the%20gonadotropin-releasing%20hormone%20analogue%20degarelix%20%28DGX%29%20for%20pharmacological%20puberty%20suppression.%20After%204%20weeks%2C%20E2%20was%20administered%20at%20different%20doses%2C%20and%20bone%20properties%20were%20analyzed%20after%20an%20additional%208%20weeks.%20DGX%20treatment%20effectively%20suppressed%20sex%20steroid%20signaling%2C%20leading%20to%20reduced%20bone%20mass%20and%20strength%2C%20which%20were%20dose-dependently%20restored%20by%20E2.%20While%20high%20E2%20doses%20resulted%20in%20an%20excessive%20increase%20in%20bone%20mass%2C%20thereby%20precluding%20further%20mechanistic%20study%2C%20lower%20doses%20resulted%20in%20a%20bone%20phenotype%20resembling%20that%20of%20native%20females.%20At%20the%20cellular%20level%2C%20DGX-mediated%20puberty%20suppression%20resulted%20in%20increased%20bone%20resorption%20that%20exceeded%20bone%20formation%2C%20and%20also%20caused%20an%20accumulation%20of%20marrow%20adipocytes.%20Subsequent%20low-dose%20E2%20administration%20reduced%20bone%20resorption%2C%20stimulated%20bone%20formation%2C%20and%20prevented%20the%20increase%20in%20bone%20marrow%20adiposity.In%20summary%2C%20we%20established%20and%20validated%20a%20mouse%20model%20that%20accurately%20mimics%20gender-affirming%20therapy%20initiated%20during%20early%20puberty%20and%20enables%20the%20study%20of%20its%20effects%20on%20bone.As%20more%20transgender%20youth%20seek%20gender-affirming%20care%2C%20it%20is%20crucial%20to%20understand%20how%20these%20medical%20interventions%20affect%20long-term%20health.%20This%20is%20especially%20true%20for%20the%20skeleton%2C%20which%20is%20still%20growing%20during%20adolescence%20and%20therefore%20very%20likely%20to%20respond%20to%20the%20introduced%20hormonal%20fluctuations.%20In%20young%20transgirls%2C%20treatment%20typically%20starts%20with%20medication%20to%20suppress%20puberty%2C%20followed%20by%20estrogen%20administration%20to%20align%20with%20their%20gender%20identity.%20However%2C%20how%20these%20interventions%20affect%20bone%20properties%20is%20not%20fully%20understood%2C%20especially%20on%20the%20long%20run.%20To%20explore%20this%20at%20a%20mechanistic%20level%2C%20we%20developed%20a%20mouse%20model%20that%20closely%20mimics%20the%20timing%20and%20effects%20of%20gender-affirming%20treatments%20used%20in%20transgirls.%20We%20found%20that%20puberty%20suppression%20led%20to%20decreased%20bone%20mass%20and%20strength%2C%20which%20was%20related%20to%20changes%20in%20bone-forming%20osteoblasts%20and%20bone-resorbing%20osteoclasts%2C%20together%20with%20increased%20fat%20content%20in%20the%20bone%20marrow.%20Subsequent%20estrogen%20administration%20dose-dependently%20reversed%20these%20effects%2C%20with%20lower%20doses%20restoring%20skeletal%20parameters%20without%20causing%20abnormally%20high%20bone%20mass%2C%20while%20higher%20doses%20led%20to%20very%20dense%20bone%20which%20does%20not%20align%20with%20clinical%20observations.%20Collectively%2C%20our%20work%20established%20a%20useful%20model%20to%20study%20how%20gender-affirming%20treatments%20influence%20growing%20bones%20and%20offers%20insights%20that%20eventually%20guide%20future%20clinical%20care%20in%20transgender%20adolescents.%22%2C%22date%22%3A%222026-01-16%22%2C%22section%22%3A%22%22%2C%22partNumber%22%3A%22%22%2C%22partTitle%22%3A%22%22%2C%22DOI%22%3A%2210.1093%5C%2Fjbmr%5C%2Fzjag009%22%2C%22citationKey%22%3A%22%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fdoi.org%5C%2F10.1093%5C%2Fjbmr%5C%2Fzjag009%22%2C%22PMID%22%3A%22%22%2C%22PMCID%22%3A%22%22%2C%22ISSN%22%3A%220884-0431%22%2C%22language%22%3A%22%22%2C%22collections%22%3A%5B%22CI4G83I
